A peptide derived from a neural cell adhesion molecule studied in animals for memory, plasticity, and neuroprotection.
Overview
FGL is a peptide fragment of the neural cell adhesion molecule that mimics NCAM's interaction with the FGF receptor. Given after training in rats it produced long-lasting memory improvement and promoted synapse formation through FGFR1. It was also neuroprotective after ischemic injury both in cultured neurons and in vivo, and systemic dosing reduced amyloid-related pathology and cognitive impairment in a rat model using the Abeta25-35 fragment. That combination of memory, plasticity and neuroprotection findings is why it attracted interest in aging and neurodegeneration. All of the evidence is from animals: human evidence is minimal, so it is experimental and not approved.
Mechanism of action
Activates the FGF receptor via an NCAM-mimicking sequence, promoting synaptic plasticity and neuronal survival.
