Trodusquemine

An aminosterol PTP1B inhibitor from dogfish shark that regenerated heart tissue in zebrafish and mice and caused fat-specific weight loss in obese mice; early human trials never produced published efficacy.

Overview

Trodusquemine blocks protein tyrosine phosphatase 1B, a brake on insulin and leptin signaling, and crosses into the brain. Animal studies showed weight loss confined to fat and regeneration of heart and other tissues after injury. Human Phase 1 work in obesity and oncology occurred years ago without published efficacy, and the compound now circulates as a research chemical with no human dosing or safety data.

Mechanism of action

Allosteric, non-competitive PTP1B inhibition enhancing insulin and leptin signaling; regenerative effects via PTP1B-dependent pathways in injured tissue.
