An oral AMPK activator with one 28-day human pilot in type 2 diabetes and recent mouse data, now sold as a research chemical and promoted as an exercise mimetic or metformin replacement.

Overview

ATX-304 keeps AMPK switched on by blocking its dephosphorylation rather than by draining cellular energy. A 28-day Phase 2a in people with type 2 diabetes already on metformin improved glucose handling and microvascular blood flow, and 2025 mouse work showed benefits in fatty liver and oxidative stress. Beyond that there is no efficacy trial, no published half-life and no long-term safety data. Its 2026 popularity followed an influencer write-up and a vendor catalog launch.

Mechanism of action

Pan-AMPK activation via inhibition of PP2C-mediated dephosphorylation at Thr172, with mild mitochondrial uncoupling.
