An oral AMPK activator with one 28-day human pilot in type 2 diabetes and recent mouse data, now sold as a research chemical and promoted as an exercise mimetic or metformin replacement.
ATX-304 keeps AMPK switched on by blocking its dephosphorylation rather than by draining cellular energy. A 28-day Phase 2a in people with type 2 diabetes already on metformin improved glucose handling and microvascular blood flow, and 2025 mouse work showed benefits in fatty liver and oxidative stress.
Mechanism: Pan-AMPK activation via inhibition of PP2C-mediated dephosphorylation at Thr172, with mild mitochondrial uncoupling.
